To create a new flag in Evernote for Windows, simply left-click in the desired part of the note, hold down the Alt key, or press Ctrl + Shift + C. In the case of the Mac client, you can simply click on the corresponding button on the toolbar.

Check and uncheck the notes with the lists can also be in Evernote for iPhone, well, in the rest of the clients the flags are only available for display.
